Amélioration génétique et utilisation des aliments à base de végétaux en pisciculture ArchiMer
Le Boucher, Richard; Dupont-nivet, M.; Laureau, S.; Labbe, Laurent; Geurden, I.; Medale, F.; Chatain, Beatrice; Vandeputte, Marc; Quillet, E..
World fish farm production is growing rapidly. However, the supply of fishmeal and fish oil, the major ingredients of fish diets, has reached a limit because of the limits in the exploitation of wild fish stocks that cannot be increased to meet a growing demand. The main response, especially in Europe, has been the progressive replacement of fishmeal and fish oil by plant products. However, the strong reduction of growth and sometimes survival of fish fed highly substituted diets has urged research on the genetic potential for adaptation to plant-based diets. Recent results clearly demonstrate that fish can be selected for their capacity to grow when fed plant-based diets, even though the underlying mechanisms remain unknown. Genotype-diet interactions...

Cofeeding of phospholipids to turbot <i>Scophthalmus maximus</i> L. larvae as a tool to reduce live food consumption OMA
Dhert, P.; González Félix, M.; Van Ryckeghem, K.; Geurden, I.; Thysen, F.; Lebegue, E.; Lavens, P.; Sorgeloos, P..
The possible benefits of feeding a formulated diet, together with <i>Artemia</i> , to first-feeding turbot larvae were investigated. Five dietary regimes were used: control ( <i>Artemia</i> only), cofeeding with a diet rich in phospholipid (PL) from bovine brain, cofeeding with a diet rich in free fatty acids (FFA) from the same source, cofeeding with a commercial diet (Lansy A2), and a formulated feed only (PL-rich diet). All treatments with artificial diets and the control treatment that received <i>Artemia</i> exclusively gave a similar survival rate of the fish at day 29. Cofeeding with artificial diets resulted in a lower growth rate than the control treatment. Cofeeding the larvae with PL-rich or FFA-rich diets did...

Differential gene expression after total replacement of dietary fish meal and fish oil by plant products in rainbow trout (Oncorhynchus mykiss) liver ArchiMer
Panserat, S.; Hortopan, G. A.; Plagnes-juan, E.; Kolditz, C.; Lansard, M.; Skiba-cassy, S.; Esquerre, D.; Geurden, I.; Medale, Francoise; Kaushik, Sadasivam; Corraze, Genevieve.
Our objective was to analyse the hepatic transcriptomes of juvenile rainbow trout fed with a plant-based diet. We focused our analysis on the total replacement of fish meal (FM) and fish oil (FO) by a 100% plant-based diet (0% FM, 0% FO). We analysed the postprandial hepatic transcriptomes of rainbow trout fed the two diets 8 h after feeding. Six total hepatic RNAs from each dietary group were hybridised against a trout cDNA microarray (9K). After treatment of the data respecting the standard MIAME (Minimum Information About a Microarray Experiment) protocol, we found that 176 hepatic genes were differentially expressed between fish fed the two diets: 96 and 80 were over-expressed and under-expressed, respectively, in trout fed the plant-based diet. A...

Effect of a dietary phospholipid supplementation on growth and fatty acid composition of European sea bass (<i>Dicentrarchus labrax</i> L.) and turbot (<i>Scophthalmus maximus</i> L.) juveniles from weaning onwards OMA
Geurden, I.; Coutteau, P.; Sorgeloos, P..
Two 40-day feeding trials using extruded diets were conducted to assess the effect of a dietary phospholipid (PL) supplementation on growth, survival and fatty acid composition of European sea bass (<i>Dicentrarchus labrax</i>) and turbot (<i>Scophthalmus maximus</i>) from weaning onwards. Two dietary treatments (FO and PL) were tested; both had an identical extruded basis (92,5% total diet weight) coated with a different lipid fraction (7,5% total diet weight). Diet PL contained 2% egg yolk PL (69% pure). In diet FO the PL was replaced by hydrogenated coconut oil. The isolipidic diets contained an equal amount of fish oil ethyl esters providing 1,6% (% diet weight) of n-3 highly unsaturated fatty acids (HUFA). A diet water...

Enrichment strategies for <i>Artemia</i> using emulsions providing different levels of n-3 highly unsaturated fatty acids OMA
Han, K.; Geurden, I.; Sorgeloos, P..
Three separate enrichment trials compared the <i>n</i>-3 highly unsaturated fatty acid (HUFA) levels in <i>Artemia franciscana</i> nauplii (Great Salt Lake, USA) as a function of the <i>n</i>-3 HUFA level in the emulsion (30% and 50%), the enrichment dose (0.05, 0.1, 0.2, 0.3 and 0.4 g1-1) and the degree of dilution of a high HUFA emulsion with an <i>n</i> -3 HUFA-free emulsion (100/0, 50/50, 25/75, 12.5/87.5 and 0/100, HUFA-rich/HUFA-free, w/w). The standard emulsions were distributed by the International Council for the Exploration of the Sea, Working Group on the Mass Rearing of Juvenile Fish (ICES). After the 48-h enrichment, the content of docosahexaenoic acid (DHA), eicosapentaenoic acid EPA) and total...

Incorporation of fatty acids from dietary neutral lipid in eye, brain and muscle of postlarval turbot fed diets with different types of phosphatidylcholine OMA
Geurden, I.; Reyes, O.S.; Bergot, P.; Coutteau, P.; Sorgeloos, P..
Previous results demonstrated the stimulating effect of soybean phosphatidylcholine (PC) on the utilization of dietary neutral lipid in larval and postlarval fish. The present study further investigated the effect of the degree of saturation of dietary PC on the enhancement of dietary fatty acid incorporation in lipids of turbot. Newly-weaned turbot were fed for 20 days on four isolipidic diets containing the same amount of highly unsaturated fatty acids (HUFA), presented either as neutral lipid, i.e. fish oil ethyl esters, or as polar lipid. Diet FO was a phospholipid-free control diet. Diets HPC, SPC and FPC were supplemented with 3% hydrogenated soybean PC, 3% native soybean PC and 3% marine fish roe PC, respectively. The three PC-supplemented diets...

Increased docosahexaenoic acid levels in total and polar lipid of European sea bass (<i>Dicentrarchus labrax</i>) postlarvae fed vegetable or animal phospholipids OMA
Geurden, I.; Coutteau, P.; Sorgeloos, P..
A 6-week feeding trial was conducted with 44d-old European sea bass (<i>Dicentrarchus labrax</i> L.) in order to examine the effect of various dietary phospholipid (PL) sources on the incorporation of n-3 highly unsaturated fatty acids (HUFA) in tissue lipids. From weaning onwards the fish received diets prepared by coating different lipid fractions (7.5% diet) on an extruded basal diet (92.5% diet). The two PL-free control diets contained 0.5 and 2% of an emulsifier blend, respectively. Seven other diets contained 2% PL, differing by their purity and origin (vegetable or animal). All diets were rendered isolipidic by the addition of hydrogenated coconut oil. Feeding the PL-supplemented diets, except the diet containing hydrolyzed soybean PL...

Influence of purified soybean phospholipids on early development of common carp OMA
Geurden, I.; Charlton, M.N.; Marion, D.; Bergot, P..
The influence of dietary phospholipid (PL) composition on early development was tested in a 25 day feeding trial with first-feeding common carp (<i>Cyprinus carpio</i>) larvae. A phosphatidylcholine (PC) fraction (PL: 94%, PC: 83%, PI: 1%) and a phosphatidylinositol (PI) fraction (PL: 100%, PC: 3%, PI: 64%) were prepared from soybean lecithin and added (2% on diet dry weight) to a casein-dextrin basal diet. Another diet was supplemented with 2% hen egg yolk PL (PL: 97%, PC: 68%, PI: 0%). A PL-deficient diet contained only peanut oil as lipid source and another one was supplemented with inositol. Final larval morphology was examined by an automatic image analysis system. The three PL fractions markedly improved growth in comparison with...

Relationship between dietary phospholipid classes and neutral lipid absorption in newly-weaned turbot, <i>Scophthalmus maximus</i> OMA
Geurden, I.; Bergot, P.; Schwarz, L.; Sorgeloos, P..
A 28-day feeding trial was conducted for comparing the effect of different dietary phospholipid (PL) classes on the growth of post-larval turbot and on the incorporation of dietary neutral lipid fatty acids into their body lipids. Prior to the experiment the turbot were weaned for one week on a PL-free diet. The nine experimental diets were isolipidic and contained an equal amount of highly unsaturated fatty acids in the form of ethyl esters. They differed by their PL content (0.1 or 2%) and by the PL class composition of the added soybean PL fractions. Compared to the PL-free diet, diets enriched with phosphatidylcholine (PC) resulted in a better growth, a higher triglyceride content (% body dry matter) and increased levels of docosahexaenoic acid (%...

Review on the dietary effects of phospholipids in fish and crustacean larviculture OMA
Coutteau, P.; Geurden, I.; Camara, M.R.; Bergot, P.; Sorgeloos, P..
A beneficial effect of dietary phospholipid (PL) supplementation in purified diets in terms of survival, growth, resistance to stress tests, and occurrence of deformities has been demonstrated in larval and juvenile stages of various species of fish and crustaceans. The exact determination of PL requirements in larvae is complicated due to the difficulty to bio-encapsulate PL in live prey. Furthermore, the great variety in purity and composition of the PL sources, and the experimental conditions (such as diet formulation and extent of co/prefeeding with live food) makes it difficult to compare requirements determined with artificial diets. Larval stages are extremely sensitive to a dietary PL deficiency and require higher levels of dietary PL than...
